Scientists have engineered nanoscale systems with a "nanoparticle spine" that could revolutionize pharmaceutical development, manufacturing, and testing. These new polymer molecules feature nanoscale backbone joints, or "stacks," that can be organized to form different shapes, such as spherical or flat, making them useful for chemical synthesis or testing of potential drug therapies.
The materials can be combined to yield a variety of shapes that can contain multiple types of materials in different concentrations (a combination of stacking). The materials can also be assembled to produce more structures, which can have different properties, such as solubility, and can be manipulated to form more complex structures, says Mingjie Zhang, a graduate student in materials science and engineering at the Harvard John A. Paulson School of Engineering and Applied Sciences in Cambridge, Mass., who led the study.
"We are only beginning to understand how form a structure in the bulk using different sizes and shapes," Zhang says. "However, this work offers a new method to generate novel nanoparticles by assembling individual of related structures. This holds the potential for many different applications, not just chemists, but also materials scientists, biochemists, and engineers."
Zhang, who has a joint appointment at Harvard-MIT and the Massachusetts Institute of Technology (MIT), and his collaborators have reported their result online April 13 in Nature Materials. Other authors are V. Michael Bursztein, an assistant professor of materials science and engineering at MIT; the late C. William Kastner, a professor of medicine and microbiology immunology at Harvard Medical School, and former MIT professor Joseph Lauer; and Zhongjie Hu, a recent Ph.D. graduate of MIT and now a postdoc in the Bursztein laboratory.
The team developed idea to stack nanoparticles on top of each other in a "x-shaped" array when one of them was introduced in 2010. The idea was that x-shaped nanoparticle should be chemically modified so it could store more of a particular material. The team realized that arrangement could be tuned to mimic several different shapes by adding chemical modifications.
To create the desired shape, team began by chemically modifying a single nanoparticle to produce five different types of "polymerization" molecules. These produced different shapes, but they all had two common characteristics: they were all about 5 nanometers cubed and had one-way channels that allowed the polymer to flow from a bottom top or.
